Flying from North America, flights typically arrive in the afternoon. If you can choose a direct transit flight to Osaka on the same day, you can expect to get into Osaka proper by 8pm or so. It's a pretty simple procedure and should be the normally preferred option over taking the train and making another transfer at Tokyo or Shinagawa.
