You give somebody a present for their birthday. The "for" expresses purpose. However, many presents are, in fact, given on the day that the birthday occurs, so in that case they are given on the birthday.

However, one does not do something "on a celebration", whether the celebration is for a birthday or not. One does something at a celebration, or at a party, or at an event. If there is a slight difference in significance between them, I would probably use: "My birthday is August the twenty-first" if I was giving details of my date of birth to an official. "My birthday is on August the twenty-first" if I was inviting friends to my party.
